A soluble CYP51 ortholog from Mycobacterium tuberculosis (MTCYP51) (Aoyama et al., 1998) has three features that make it an attractive model for developing a screening assay and defining binding of positive hits. Apparently, other than the phenyl ring functionalities attached to the carbon chirality center may also be explored. At the same time, the N-[4-pyridyl]-formamide bulk could serve as universal building block in design of species-specific CYP51 inhibitors. It is apparent that functional groups other than phenyl rings attached to the carbon chirality center may also be explored. Further, the N-[4-pyridyl]-formamide bulk could serve as a universal building block in design of species-specific CYP51 inhibitors.

Kitchener et al. (2005) studied interstate variability on all programs providing personal care, including both 1915(c) waivers and PCS programs, and documented a continuation of variation between the provision and implementation of PCS, as documented in an earlier study by Muramatsu and Campbell (2002). State-to-state variability in the provision and implementation of PCS is significant, however (Kitchener et al. 2005).

Giardia lamblia, a flagellated protozoan parasite, causes gastrointestinal disease known as giardiasis in humans and is responsible for major waterborne outbreaks of diarrhea in the United States. Due to early divergence during eukaryotic evolution, Giardia's translation machinery deviates from that of the higher eukaryotes. In higher eukaryotes, translation initiation involves a scanning mechanism wherein the ribosome bound to the 5'end of the capped messenger RNA (mRNA) scans in the 5' to 3' direction along the long 5'UTR in search of an initiation codon. In contrast, mRNAs of Giardia possess very short 5'untranslated regions (5'UTRs) and translation initiation... Giardia lamblia, a flagellated protozoan parasite, causes the gastrointestinal disease giardiasis in humans and is responsible for major waterborne outbreaks of diarrhea in the United States. In addition to its importance as a pathogen, Giardia is also significant as a model organism that gives understanding of basic biological processes. Due to its early divergence during eukaryotic evolution, Giardia possesses simplified translational machinery that provides a tractable and productive system for studying the details of translation. Translation initiation in Giardia...

Objectives: Sterol 14 a-demethylase (CYP51) is a key target in treatment of infectious microbes due to its involvement in biosynthesis of membrane sterols. As a cytochrome P450, CYP51 has spectral properties making it a convenient target for high throughput screening (HTS) technologies via monitoring changes in optical spectra in response to ligand binding.
Methods: We screened a library of 20,000 organic molecules against Mycobacterium tuberculosis CYP51...

Polo-like kinase (Plk) plays multiple roles in mitosis and cytokinesis among all eukaryotes examined thus far. Its down-regulation interferes with spindle formation during mitosis leading to cell cycle arrest and apoptosis. Here we report that in an ancient eukaryote Trypanosoma brucei the only Plk homolog, TbPlk, controls only cytokinesis. This is regulated primarily by the kinetoplast cycle instead of the nuclear cycle. A knockdown of the expression of TbPlk by RNA interference in the procyclic form of T. brucei resulted in... Polo-like kinase (Plk) typically plays multiple roles in both mitosis and cytokinesis in eukaryotes. We report that TbPlk, the only Plk homolog in the ancient eukaryote Trypanosoma brucei, controls cytokinesis but not mitosis. In T. brucei mitosis is regulated by the kinetoplast cycle, not the nuclear cycle. We used RNAi to knock down TbPlk expression in the procyclic form of T. brucei.
